Malcolm, the European silver medallist, clocked 20.75 seconds while Spearmon streaked to victory in an impressive 19.95 secs.
David Oliver powered an expected victory in the 110m hurdles but Andy Turner warmed up for next month's Commonwealth Games when he finished second.
European champion Turner crossed the line in 13.48 secs, two tenths off his time in Barcelona.
Blanka Vlasic delighted an adoring home crowd as she sailed over 2.05 metres, equalling her season's best, to win the high jump.
She made three attempts to clear 2.10m, a centimetre higher than the 23-year-old world mark of Bulgaria's Stefka Kostadion, but never really got close.
"You can't imagine what this means to me," said a tearful and emotional Vlasic.
"It a moment that my family and I will never forget. I'm so proud to be part of it, and so proud that the crowd supported everybody, in all the events. It was just fantastic."
Michael Bingham and Marytn Rooney were part of a powerful European team in the concluding 4x400m relay - which included new European champion Kevin Borlee.
However, the quartet were still outclassed by a powerful Americas team, starring Bershawn Jackson and Ricardo Chambers, who clocked 2.59.00 to break a 29-year old meeting record.
Despite that defeat the European team still won the title, overhauling the overnight lead held by the Americas.
They amassed 429 points, while the Americas were just behind with 419.5 points.
