Murray is the reigning champion of the Masters 1000 event but won his title indoors last October.
The new world number three faces a potential third round encounter for France's Tommy Robredo while world number five Juan Del Potro and world number two Roger Federer also lurk in his half of the draw.
Murray holds a 3-1 winning advantage over Chela but they have never played on the Argentines favoured surface.
He has played Bolelli just once - beating him in Madrid last year on the way to the title.
Meanwhile, Anne Keothavong will face Czech Republic's Lucie Safarova after winning her first round match in Madrid against Colombia's Mariana Duque Marino.
The world number 53 was made to work by Marino, placed 71 places below her on the WTA rankings, before advancing 6-3, 7-6.
Safarova beat the British number one 6-4, 7-5 in their last encounter at the Miami Masters earlier this year.
